The possible ameliorative effect of Resveratrol on the lead exposed parotid glands in albino rats: Insights into glandular histology and DNA damage

Amira A.r. Moawad.

Abstract
Background and Aims:
Exposure of the public to lead is considered a hazard to the health due to its lifelong harmful effects. Considering the incorporation of antioxidants as it is considered a successful treatment option, resveratrol has been suggested to forestall the harmful effect induced by exposure to lead. The goal of this study is to investigate the histological, immunohistochemical and DNA variations that might evolve in the tubuloacinar system of the parotid salivary glands of adult albino rats following ingestion of lead acetate, and to recognize the possible protective outcome of resveratrol.
Methods:
The investigation sample composed of 45 male Wistar rat. Group I were obtained 50 ml/kg/day of distilled water for fifty-five days. Group II were received 50 mg/kg/day of lead (Pb) acetate for fifty-five days. Group III were received 50 mg/kg/day of lead acetate for fifty-five days and simultaneously, 10 mg/kg/day of resveratrol (RSV) for 28 days. Upon ending the investigation, the rats were terminated, and their parotid glands were cut up and handled for H&E stain, caspase 3, and comet assay evaluation.
Results:
The microscopic detection in Group II revealed noticeable deteriorating alterations in the parotid gland. Though, in Groups III, remarkable renewal was noticed subsequently to injection with resveratrol. The resveratrol group exhibited considerable declination in DNA deteriorations in the parotid gland proportional to the lead group and almost equivalent to control.
Conclusion:
Resveratrol showed noticeable potency in renewal the parotid salivary glands of rats exposed to lead..

Key words: lead; resveratrol; parotid gland; Caspase 3; comet assay
